Background: Flavin-dependent monooxygenases are involved in key biological processes as they catalyze a wide variety of chemo-, regio- and enantioselective oxygenation reactions. Flavoprotein monooxygenases are frequently encountered in micro-organisms, most of which require further functional and biocatalytic assessment. Here we investigated the function of the AbMak1 gene, which encodes a group A flavin monooxygenase in the plant pathogenic fungus Alternaria brassicicola, by generating a deficient mutant and examining its phenotype. Acidic polysaccharides from Psidium cattleianum (Araçá) BABT

The aim of this work was to study the acidic polysaccharides of Psidium cattleianum. Pectic fractions were obtained by successive extractions with water, EDTA and sodium carbonate (4 and 25ºC). Monosaccharide composition, total carbohydrates, uronic acids and protein contents were determined for each fraction. The yield of water and EDTA-soluble fractions and high content of uronic acid were consistent with the presence of pectins probably arising from the middle lamellae. On the other hand, sodium carbonate-soluble pectins had a higher neutral sugar content, indicating highly branched polysaccharides in these fractions, consistent with pectins from cell wall.

According to the model frequently used as reference, plant cell growth occurs only when turgor surpasses a threshold. This model was proposed considering a cylindrical cell of constant wall thickness immerged in a water solution, with viscoelastic behaviour, hydraulic conductivity, variable extensibility, and unidirectional elongation. The author, Lockhart (1965), did not consider the effects of apoplasm hydrostatic potential, a subject treated later by Calbo and Pessoa (1994) who argued that this component of cell potential would interfere with cell growth rate. The experiment was conducted to evaluate the bromatological characteristics and the in vitro digestibility of four sugarcane varieties, subjected or not to hydrolysis, with quicklime. A completely randomized design was employed with three replications arranged in a 4 × 2 factorial scheme, with four sugarcane varieties (SP 52454, RB 867515, RB 855536 and IAC 862480), hydrolyzed or not. There was significant effect on brix (p < 0.05) and industrial fiber (p < 0.05), and IAC 862480 variety had the lowest levels of industrial fiber. UDP-glucose dehydrogenase (UGDH) catalyzes the oxidation of UDP-glucose (UDP-Glc) to UDP-glucuronate (UDP-GlcA), a key sugar nucleotide involved in the biosynthesis of plant cell wall polysaccharides. A full-length cDNA fragment coding for UGDH was cloned from the cambial region of 6-month-old E. grandis saplings by RT-PCR. The 1443-bp-ORF encodes a protein of 480 amino acids with a predicted molecular weight of 53 kDa. The recombinant protein expressed in Escherichia coli catalyzed the conversion of UDP-Glc to UDP-GlcA, confirming that the cloned cDNA encodes UGDH. This review summarises recent advances in the chemistry, physiology and molecular properties of coffee carbohydrates with a particular focus on the cell wall polysaccharides. The results of detailed chemical studies have demonstrated novel structural features of both the galactomannans and the arabinogalactan polysaccharides of the green and roasted coffee bean. Cryptococcus neoformans is an important opportunistic fungal pathogen that causes life-threatening infection of the central nervous system. A major virulence factor for C. neoformans is the production of melanin in the cell wall. Using transmission electron microscopy, we studied the cell walls of three pairs of isolates obtained from patients with dual cryptococcal infections, where a melanotic and an albino strain were isolated from the CSF of each patient. ß-Glucans are soluble fibers with physiological functions, such as interference with absorption of sugars and reduction of serum lipid levels. The objective of the present study was to analyze the distribution of ß-glucans in different tissues of the African grass species Rhynchelytrum repens and also to evaluate their hypoglycemic activity. Leaf blades, sheaths, stems, and young leaves of R. repens were submitted to extraction with 4 M KOH. Analysis of the fractions revealed the presence of arabinose, glucose, xylose, and traces of rhamnose and galactose. The use of monoclonal antibodies for specific pectic epitopes is an important tool in the study of the cell wall. Throughout the development of mucilage cells of Araucaria angustifolia (Bertol.) Kuntze, a gradient of distribution was observed in relation to the pectic de-esterification, as well as to the increase of galactan and arabinan epitope distribution, and to the reduction of arabinogalactans proteins (AGPs) epitope at maturity. AGP and methyl-esterified homogalacturonan (HGA) were present in the mucilage. Galactans and arabinans were also observed in the mucilage, though with weak labelling. Immunomodulatory properties of cell wall extract from Enterococcus faecalis CECT7121 BJID

The aim of this study was to investigate the immunomodulatory properties of cell wall extract from Enterococcus faecalis CECT7121, measuring the induction of cytokines TNF-α, IL-6, IL-10 and IL-12 from human peripheral blood mononuclear cells (PBMCs). Cell wall extract was prepared from their growth in brain heart infusion broth (18 h, 35 °C). Subsequently, toxicity of the obtained cell wall extract was tested in Balb-C mice. ABSTRACT The aim of this study was to investigate anatomical characteristics of barley (Hordeum vulgare L.) internodes and to reveal the connection with activity of guaiacol-peroxidases, their isoenzyme profiles, lignin, and cellulose content. Three lower internodes were sampled at elongation stage and differed in their anatomical features by showing that lignin deposition follows developmental pattern. Epidermis and hypodermis in the first internode were characterized by continuous layer of the lignified cells while in the second and third internode thin walled parenchyma cells could be observed in the hypodermis. Occurrence of xyloglucan containing protuberances in the storage cell walls of cotyledons of Hymenaea courbaril L. Rev. Bras. Bot.

Despite the suggestions of its pectic composition, no clear evidence for this has been presented. Here we show the occurrence of such a structure in walls of cells from cotyledons of Hymenaea courbariI L. These cells are known to accumulate large amounts of storage xyloglucan in the wall and, in this case, the protuberances seem to contain this storage polysaccharide rather than pectin. A hypothetical sequence of events leading from wall strands to protuberances was assembled based on scanning electron microscopy observations. On this basis, a tentative model for how polysaccharides are distributed into the wall, near the regions where protuberances are found, is proposed to explain the presence of storage xyloglucan in their composition.

In our studies to isolate the genes the lignin biosynthesis in S. spontaneum we have had great difficulty in RT-PCR reactions. Yeasts discarded in industrial processes can be used as a nutritional supplement and to extract cellular components with biotechnological aims. In this study, the humoral immune response of Swiss mice treated with mannoproteins (MP) from the yeast Saccharomyces uvarum was evaluated. The mice were treated with MPs at different doses and times and inoculated with 2% sheep red blood cells. An increase in total Ig in mice treated with 100 μg of MP at the time of immunization or 24 h before was observed in the primary immune response; in the secondary immune response, an increase was observed in total Ig values for all groups, and an increase of IgG was observed in the mice treated with MPs (100 μg) at the time of immunization or 24 h before. For a long time sporotrichosis has been regarded to have a low incidence in Brazil; however, recent studies demonstrate that not only the number of reported cases but also the incidence of more severe or atypical clinical forms of the disease are increasing. Recent data indicate that these more severe forms occur in about 10% of patients with confirmed diagnosis. The less frequent forms, mainly osteoarticular sporotrichosis, might be associated both with patient immunodepression and zoonotic transmission of the disease. The extracutaneous form and the atypical forms are a challenge to a newly developed serological test, introduced as an auxiliary tool for the diagnosis of unusual clinical forms of sporotrichosis.
